NM_177438.3(DICER1):c.412T>G (p.Trp138Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W138G variant (also known as c.412T>G), located in coding exon 3 of the DICER1 gene, results from a T to G substitution at nucleotide position 412. The tryptophan at codon 138 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
