This variant lies in the SBDS gene (transcript NM_016038.4) at coding-DNA position 40, where A is replaced by T; at the protein level this means replaces asparagine at residue 14 with tyrosine — a missense variant. Submitter rationale: The p.N14Y variant (also known as c.40A>T), located in coding exon 1 of the SBDS gene, results from an A to T substitution at nucleotide position 40. The asparagine at codon 14 is replaced by tyrosine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species.
