NM_003001.5(SDHC):c.409T>G (p.Trp137Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W137G variant (also known as c.409T>G), located in coding exon 6 of the SDHC gene, results from a T to G substitution at nucleotide position 409. The tryptophan at codon 137 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
