Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.4091C>G (p.Ser1364Cys), citing Ambry Variant Classification Scheme 2023: The p.S1364C variant (also known as c.4091C>G), located in coding exon 33 of the TSC2 gene, results from a C to G substitution at nucleotide position 4091. The serine at codon 1364 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
