Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001844.5(COL2A1):c.1222-2A>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects an acceptor splice site in intron 19 of the COL2A1 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with Stickler syndrome (PMID: 8737653). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 17374). Studies have shown that disruption of this splice site results in activation of a cryptic splice site and introduces a premature termination codon (PMID: 8737653). The resulting mRNA is expected to undergo nonsense-mediated decay. For these reasons, this variant has been classified as Pathogenic.
