Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.403G>T (p.Asp135Tyr), citing Ambry Variant Classification Scheme 2023: The p.D135Y variant (also known as c.403G>T), located in coding exon 5 of the RAD54L gene, results from a G to T substitution at nucleotide position 403. The aspartic acid at codon 135 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
