NM_005359.6(SMAD4):c.400G>C (p.Glu134Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome; Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E134Q variant (also known as c.400G>C), located in coding exon 2 of the SMAD4 gene, results from a G to C substitution at nucleotide position 400. The glutamic acid at codon 134 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
