Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.3995T>C (p.Leu1332Pro), citing Ambry Variant Classification Scheme 2023: The p.L1332P variant (also known as c.3995T>C), located in coding exon 30 of the NF1 gene, results from a T to C substitution at nucleotide position 3995. The leucine at codon 1332 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
