NM_004415.4(DSP):c.397C>G (p.Gln133Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q133E variant (also known as c.397C>G), located in coding exon 3 of the DSP gene, results from a C to G substitution at nucleotide position 397. The glutamine at codon 133 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
