Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.3967T>C (p.Ser1323Pro), citing Ambry Variant Classification Scheme 2023: The p.S1323P variant (also known as c.3967T>C), located in coding exon 20 of the BLM gene, results from a T to C substitution at nucleotide position 3967. The serine at codon 1323 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
