NM_001365951.3(KIF1B):c.4105C>G (p.Leu1369Val) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L1323V variant (also known as c.3967C>G), located in coding exon 36 of the KIF1B gene, results from a C to G substitution at nucleotide position 3967. The leucine at codon 1323 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
