NM_003072.5(SMARCA4):c.3912G>C (p.Gln1304His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q1304H variant (also known as c.3912G>C), located in coding exon 27 of the SMARCA4 gene, results from a G to C substitution at nucleotide position 3912. The glutamine at codon 1304 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
