Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.1013A>G (p.His338Arg), citing Ambry Variant Classification Scheme 2023: The p.H338R variant (also known as c.1013A>G), located in coding exon 3 of the AXIN2 gene, results from an A to G substitution at nucleotide position 1013. The histidine at codon 338 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
