Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.388C>T (p.His130Tyr), citing Ambry Variant Classification Scheme 2023: The p.H130Y variant (also known as c.388C>T), located in coding exon 4 of the NF1 gene, results from a C to T substitution at nucleotide position 388. The histidine at codon 130 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
