NM_017617.5(NOTCH1):c.3870C>G (p.Asp1290Glu) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1290E variant (also known as c.3870C>G), located in coding exon 23 of the NOTCH1 gene, results from a C to G substitution at nucleotide position 3870. The aspartic acid at codon 1290 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
