NM_007194.4(CHEK2):c.383C>G (p.Pro128Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P128R variant (also known as c.383C>G), located in coding exon 2 of the CHEK2 gene, results from a C to G substitution at nucleotide position 383. The proline at codon 128 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
