Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.382G>A (p.Ala128Thr), citing Ambry Variant Classification Scheme 2023. This variant lies in the HOXB13 gene (transcript NM_006361.6) at coding-DNA position 382, where G is replaced by A; at the protein level this means replaces alanine at residue 128 with threonine — a missense variant. Submitter rationale: The p.A128T variant (also known as c.382G>A), located in coding exon 1 of the HOXB13 gene, results from a G to A substitution at nucleotide position 382. The alanine at codon 128 is replaced by threonine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
