This variant lies in the TNNI3 gene (transcript NM_000363.5) at coding-DNA position 380, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 127 with glycine — a missense variant. Submitter rationale: The p.D127G variant (also known as c.380A>G), located in coding exon 7 of the TNNI3 gene, results from an A to G substitution at nucleotide position 380. The aspartic acid at codon 127 is replaced by glycine, an amino acid with similar properties. An alternate amino acid substitution at this codon, p.D127Y, has been reported as de novo in an early onset restrictive cardiomyopathy case (Klauke B et al. PLoS ONE, 2017 Dec;12:e0189489). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
