NM_001089.3(ABCA3):c.3805G>C (p.Glu1269Gln) was classified as Uncertain significance for Hereditary pulmonary alveolar proteinosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1269Q variant (also known as c.3805G>C), located in coding exon 22 of the ABCA3 gene, results from a G to C substitution at nucleotide position 3805. The glutamic acid at codon 1269 is replaced by glutamine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6498 samples (12996 alleles) with coverage at this position. This amino acid position is well conserved in available vertebrate species; however, glutamine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
