Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001903.5(CTNNA1):c.37T>C (p.Trp13Arg), citing Ambry Variant Classification Scheme 2023: The p.W13R variant (also known as c.37T>C), located in coding exon 1 of the CTNNA1 gene, results from a T to C substitution at nucleotide position 37. The tryptophan at codon 13 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
