This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 377, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 126 with glycine — a missense variant. Submitter rationale: The p.D126G variant (also known as c.377A>G), located in coding exon 4 of the ATM gene, results from an A to G substitution at nucleotide position 377. The aspartic acid at codon 126 is replaced by glycine, an amino acid with similar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
