Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_017849.4(TMEM127):c.376A>G (p.Thr126Ala), citing Ambry Variant Classification Scheme 2023: The p.T126A variant (also known as c.376A>G), located in coding exon 2 of the TMEM127 gene, results from an A to G substitution at nucleotide position 376. The threonine at codon 126 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
