NM_000257.4(MYH7):c.374T>A (p.Val125Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V125D variant (also known as c.374T>A), located in coding exon 3 of the MYH7 gene, results from a T to A substitution at nucleotide position 374. The valine at codon 125 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
