Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001148.6(ANK2):c.3730A>G (p.Ser1244Gly), citing Ambry Variant Classification Scheme 2023: The p.S1244G variant (also known as c.3730A>G), located in coding exon 31 of the ANK2 gene, results from an A to G substitution at nucleotide position 3730. The serine at codon 1244 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
