Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000363.5(TNNI3):c.372+2T>A, citing Ambry Variant Classification Scheme 2023: The c.372+2T>A intronic variant results from a T to A substitution two nucleotides after coding exon 6 in the TNNI3 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and will result in the creation or strengthening of a novel splice donor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function of TNNI3 has not been clearly established as a mechanism of disease. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
