Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001184.4(ATR):c.3704A>G (p.His1235Arg), citing Ambry Variant Classification Scheme 2023: The p.H1235R variant (also known as c.3704A>G), located in coding exon 19 of the ATR gene, results from an A to G substitution at nucleotide position 3704. The histidine at codon 1235 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
