Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.3838T>C (p.Cys1280Arg), citing Ambry Variant Classification Scheme 2023: The p.C1234R variant (also known as c.3700T>C), located in coding exon 33 of the KIF1B gene, results from a T to C substitution at nucleotide position 3700. The cysteine at codon 1234 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
