Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.3691T>G (p.Leu1231Val), citing Ambry Variant Classification Scheme 2023: The p.L1231V variant (also known as c.3691T>G), located in coding exon 24 of the ATM gene, results from a T to G substitution at nucleotide position 3691. The leucine at codon 1231 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
