Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000371.4(TTR):c.367C>G (p.Arg123Gly), citing Ambry Variant Classification Scheme 2023: The p.R123G variant (also known as c.367C>G), located in coding exon 4 of the TTR gene, results from a C to G substitution at nucleotide position 367. The arginine at codon 123 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
