Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.3675G>A (p.Met1225Ile), citing Ambry Variant Classification Scheme 2023: The p.M1225I variant (also known as c.3675G>A), located in coding exon 27 of the NF1 gene, results from a G to A substitution at nucleotide position 3675. The methionine at codon 1225 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
