Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_014845.6(FIG4):c.365A>G (p.His122Arg), citing Ambry Variant Classification Scheme 2023: The p.H122R variant (also known as c.365A>G), located in coding exon 4 of the FIG4 gene, results from an A to G substitution at nucleotide position 365. The histidine at codon 122 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
