Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000321.3(RB1):c.364A>C (p.Lys122Gln), citing Ambry Variant Classification Scheme 2023: The p.K122Q variant (also known as c.364A>C), located in coding exon 3 of the RB1 gene, results from an A to C substitution at nucleotide position 364. The lysine at codon 122 is replaced by glutamine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
