Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.3641A>C (p.Glu1214Ala), citing Ambry Variant Classification Scheme 2023: The p.E1215A variant (also known as c.3644A>C), located in coding exon 8 of the ALMS1 gene, results from an A to C substitution at nucleotide position 3644. The glutamic acid at codon 1215 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
