Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_013372.7(GREM1):c.363G>T (p.Gln121His), citing Ambry Variant Classification Scheme 2023: The p.Q121H variant (also known as c.363G>T), located in coding exon 1 of the GREM1 gene, results from a G to T substitution at nucleotide position 363. The glutamine at codon 121 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
