Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.3637A>G (p.Lys1213Glu), citing Ambry Variant Classification Scheme 2023: The p.K1213E variant (also known as c.3637A>G), located in coding exon 22 of the CFTR gene, results from an A to G substitution at nucleotide position 3637. The lysine at codon 1213 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
