NM_000051.4(ATM):c.361T>G (p.Leu121Val) was classified as Uncertain significance for Familial cancer of breast by KCCC/NGS Laboratory, Kuwait Cancer Control Center, citing ACMG Guidelines, 2015: The p.L121V variant , located in coding exon 5 of the ATM gene, results from a T to G substitution at nucleotide position 361. The leucine at codon 121 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved . ClinVar database contain an entry (ID:1733317) by single clinical laboratory classified as variant of uncertain significance . In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
