NM_000093.5(COL5A1):c.3607C>G (p.Arg1203Gly) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1203G variant (also known as c.3607C>G), located in coding exon 46 of the COL5A1 gene, results from a C to G substitution at nucleotide position 3607. The arginine at codon 1203 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
