Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_006258.4(PRKG1):c.1148T>C (p.Val383Ala), citing Ambry Variant Classification Scheme 2023: The p.V383A variant (also known as c.1148T>C), located in coding exon 10 of the PRKG1 gene, results from a T to C substitution at nucleotide position 1148. The valine at codon 383 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
