This variant lies in the DNAH5 gene (transcript NM_001369.3) at coding-DNA position 358, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 120 with asparagine — a missense variant. Submitter rationale: The p.D120N variant (also known as c.358G>A), located in coding exon 4 of the DNAH5 gene, results from a G to A substitution at nucleotide position 358. The aspartic acid at codon 120 is replaced by asparagine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
