Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_020822.3(KCNT1):c.3583A>C (p.Ile1195Leu), citing Ambry Variant Classification Scheme 2023: The p.I1195L variant (also known as c.3583A>C), located in coding exon 30 of the KCNT1 gene, results from an A to C substitution at nucleotide position 3583. The isoleucine at codon 1195 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
