Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_053025.4(MYLK):c.3578G>T (p.Ser1193Ile), citing Ambry Variant Classification Scheme 2023: The p.S1193I variant (also known as c.3578G>T), located in coding exon 17 of the MYLK gene, results from a G to T substitution at nucleotide position 3578. The serine at codon 1193 is replaced by isoleucine, an amino acid with dissimilar properties. This alteration has been reported in a thoracic aortic aneurysm and dissection (TAAD) cohort (Weerakkody R et al. Genet Med, 2018 11;20:1414-1422). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
