Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.3544C>G (p.Leu1182Val), citing Ambry Variant Classification Scheme 2023: The p.L1182V variant (also known as c.3544C>G), located in coding exon 27 of the CACNA1C gene, results from a C to G substitution at nucleotide position 3544. The leucine at codon 1182 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
