NM_006904.7(PRKDC):c.3524A>T (p.His1175Leu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H1175L variant (also known as c.3524A>T), located in coding exon 30 of the PRKDC gene, results from an A to T substitution at nucleotide position 3524. The histidine at codon 1175 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
