NM_001365951.3(KIF1B):c.3653G>A (p.Arg1218Gln) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1172Q variant (also known as c.3515G>A), located in coding exon 31 of the KIF1B gene, results from a G to A substitution at nucleotide position 3515. The arginine at codon 1172 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
