Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.3638C>A (p.Pro1213His), citing Ambry Variant Classification Scheme 2023: The p.P1167H variant (also known as c.3500C>A), located in coding exon 31 of the KIF1B gene, results from a C to A substitution at nucleotide position 3500. The proline at codon 1167 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
