This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 348, where T is replaced by A; at the protein level this means replaces aspartic acid at residue 116 with glutamic acid — a missense variant. Submitter rationale: The p.D116E variant (also known as c.348T>A), located in coding exon 2 of the MSH2 gene, results from a T to A substitution at nucleotide position 348. The aspartic acid at codon 116 is replaced by glutamic acid, an amino acid with highly similar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was reported to be functionally neutral (Jia X et al.
