Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.3487G>A (p.Val1163Ile), citing Ambry Variant Classification Scheme 2023: The p.V1163I variant (also known as c.3487G>A), located in coding exon 22 of the CFTR gene, results from a G to A substitution at nucleotide position 3487. The valine at codon 1163 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
