Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.344T>C (p.Met115Thr), citing Ambry Variant Classification Scheme 2023: The p.M115T variant (also known as c.344T>C), located in coding exon 3 of the APC gene, results from a T to C substitution at nucleotide position 344. The methionine at codon 115 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. However, this alteration is predicted to be benign and tolerated by PolyPhen and SIFT in silico analyses, respectively. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
