NM_005751.5(AKAP9):c.3449T>G (p.Leu1150Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L1150R variant (also known as c.3449T>G), located in coding exon 9 of the AKAP9 gene, results from a T to G substitution at nucleotide position 3449. The leucine at codon 1150 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
