NM_000179.3(MSH6):c.3432G>C (p.Met1144Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M1144I variant (also known as c.3432G>C), located in coding exon 5 of the MSH6 gene, results from a G to C substitution at nucleotide position 3432. The methionine at codon 1144 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
